Mouse fur mite investigation: Updates

9/28/12

To date, 220 cages of mice of various strains have been started on treatment with Ivermectin chow, with no mortality or adverse reactions noted. 105 cages have completed the entire 8 week treatment period and were released to our locked clean room, L07, to begin breeding. Due to special circumstances, we have allowed a small number of animals to be bred during treatment. Thus far, 14 litters have been born in the treatment room, most to dams that were on the medicated chow during and after delivery, and weanlings were kept on the diet until the parents were due to be released. One entire litter died at P1-3, seemingly due to maternal abandonment, however 3 more litters were born in the cage during treatment and all survived, therefore it is unlikely the medication played a role in the death of the first litter. The remainder of the litters either survived until weaned and released, or are still nursing and/or undergoing treatment, and appear normal at this time.

7/10/12

The first group of cages (about 50) were started on the medicated diet on 7/2; thus far no adverse reactions have been noted. The ventilated racks in the treatment room (L56) were all tested via PCR and found to be negative for mite DNA prior to being populated with the mice for treatment. 48 pelage tapes were collected from clinical cases and all were negative. A total of 252 clinical case pelage tapes have been collected since the end of April with only one revealing a positive result. Routine collection of tapes from all clinical cases is being discontinued.

6/22/12

Ivermectin diet is in.  We plan on starting the treatment process in the Whitehead Building for the managed breeding service (rooms L52, 54, 56, 58, 60) and rooms where mites have recently been found (L11, 21 and 48).  L56 has been cleaned and decontaminated and will serve as a pass-through treatment room.  Investigators will be asked to identify young mice that can be spared.  These mice will be treated with ivermectin in the treatment room for 8 weeks and then will be moved to a clean room to commence breeding.  Once progeny are confirmed to be viable, and in consultation with investigators, founder animals will either be euthanized or used experimentally.  In this way, our intention is that colonies will gradually be freed of mites and the number of clean room expanded while minimizing the impact to research.  Treatment is expected to start next week.  Karen Lieber is managing treatment and will contact investigators individually to arrange for transfers of mice.

6/15/12

57 clinical case and randomly collected pelage tapes were examined, all were negative. Exhaust swab PCR on the ‘test’ rack in L11 was equivocally positive at week 2 after placement.

6/1/12

50 clinical case pelage tapes were collected, all were negative. Colony tapes and pelt PCR failed to corroborate the positive pelage tape from L13. L11 had 2 of 33 tapes positive from around positive case. L21 had 1 of 8 tapes positive from around positive case. L48 had 7 of 24 tapes positive from one of two racks that had sentinels test positive, and indicate a heavy mite burden in this colony. The other positive rack had 2 of 18 tapes positive. PCR negative racks were placed in L11 and L60. The rack in L60 yielded positive PCR one week after populated, while the one in L11 remained negative; a 2 week sample has been submitted from L11.

5/11/12

54 clinical case pelage tapes were collected, one was positive (WBRB L13). Colony pelage tapes as well as PCR swabs were collected and results are pending. Tapes from cages near positive cases in L11, L13, L21 and L48 are being processed; thus far  positives have been confirmed in L11.

5/4/12

Initial surface swab PCR testing to validate procedure for assessing status of static racks yielded positive results for known positive cages. Validation testing is ongoing. PCR testing of positive dirty racks, followed by cleaning and then retesting yielded negative results, indicating the racks likely can be cleaned to remove all mite DNA. This survey will be repeated to ensure cleaning will result in negative PCR’s. 37 pelage tapes collected from clinical cases yielded no positive results.

4/27/12

We continue to collect data in attempt to assess prevalence of fur mites on campus.  General PCR survey of room results are mostly in, showing positives in the Whitehead, Woodruff, and Clinic B buildings.  Further “drilling down” to ascertain the meaning behind these findings is ongoing.  Aside from the data presented below, all clinical cases are now being assessed by pelage tape test and cage and rack cleaning techniques are also being evaluated.

4/19/12

PCR sampling at the room level of all mouse rooms on campus has been done with results pending.  A study analyzing the effectiveness of rack cleaning has also begun.  Sampling of all racks on L58 in Whitehead showed all ventilated racks to be positive and the static rack to be negative.  Validation sampling is being done on the testing methodology of static racks.

4/13/12

Evaluation for the presence of mouse fur mites on campus is ongoing.  A general survey of mouse rooms with PCR testing of racks is being conducted campus-wide.  In order to tease out the meaning of positive PCR findings, follow-up testing and “drilling down” into rooms is being done.  Results of the general survey of the lower floor of the Whitehead Building are in, with 15 rooms positive by rack PCR and 9 rooms negative.  Positive PCR results for 4 of the 15 positive rooms are supported by the finding of a mite or mite egg on pelage tape tests of clinical cases or sentinel mice.  We have also conducted pelage tape tests of at least one breeder mouse per strain per rack in rooms L58 and 60.  Approximately 130 pelage tape tests revealed one positive colony mouse in L60.

General survey conducted using rack PCRs of the two mouse rooms in Rollins was negative.

General survey of the rest of campus is ongoing.  Treatment plans will follow collection of data of the prevalence and location of the problem.  Updates will be provided here.  Data available to date in tabular form is below.
